  Comparative transcriptome analysis of two accessions of G. herbaceum L. in response to drought  

Genome-wide gene expression profiling and detailed physiological investigation were used for understanding the molecular mechanism and physiological response of G. herbaceum, which governs the adaptability of plants in drought conditions. Fourteen accessions of G. herbaceum were subjected to mannitol stress for preliminary screening; two genotypes namely Vagad and GujCot 21 are drought tolerant whereas RAHS-14 and RAHS-IPS-187 are drought sensitive. The differential expression pattern of genes among four genotypes of G. herbaceum in root and leaf tissue was analyzed under drought stress by transcriptome sequencing on Roche's 454 Genome Sequencer. A total, 318872 reads from all genotypes were obtained. The total reads were tagged and pooled to form one large dataset which was assembled into contigs by using Roche’s GS-assembler (overlap length ≥40 bp and 90% identity).
